TideCell-100 High-Density Cell Culture System (TideCell-100 System) is a fixed-bed bioreactor designed for the mass production of cell, protein and virus. The high surface-to-volume ratio leads to the significantly reduced production space. The inherent benefit of “TideCell mechanism” ensures that the process can behave as expected at the final scale without unavoidable risks (limit oxygen supply, uneven nutrient distribution, channel effect, etc). The output of cells can reach around 5×1012, which is equivalent to 2,000 L conventional stir tank bioreactor.
  

Integrated with single-use consumables and industry - recognized components, TideCell-100 System simplifies the operation, maintenance, and parts replacement, and thus fast turnaround of production campaigns can be achieved. The single-use mixing bag (500 L or 1,000 L), with bearingless, magnetic pump, ensures efficient, low-shear stress, and foam-free mass transfer. This feature, combined with a sterile medium supply chamber, help maintain pH/nutrient consistency from one cell batch to the next.

The program TideCommander-100 allows the user to set up parameters (weight, agitation, pH, DO, CO2 concentration, temperature, medium feeding, etc), and to show comparisons of various batches, trends within different periods, and pre-configured reports. The logging data / event record and alarm functions are also available with respect to the security issue, minimizing potential risks while manufacturing.
 
 
In addition, CESCO Bioengineering fully supports all necessary document packages to support validation. Documents for technical descriptions (drawings, accessory list, etc), qualification of the system (DQ, IQ, OQ, etc), standard operation procedures, traceable calibration certificate.

Product Description

 Principle:

 
TideCell HD Cell Culture System inherits the same patented principle of BelloCell bottles. Packed with porous BioNOC II matrices, the matrix vessel functions as an artificial lung with a huge contact surface for nutrition and aeration during cell culture. The culture medium is periodically drawn from the external Mixing Bag into the matrix vessel to submerge the matrices and withdrawn from the vessel to expose the matrices, driven by the applied air pressure. Thus, cells entrapped inside the matrices are able to receive sufficient nutrients and oxygen, while metabolic wastes such as lactate and carbon dioxide can be efficiently carried away in a gentle manner. The concept of the design is to solve problems (shear stress, limited oxygen supply, and low cell density) frequently occurring on the commercial models, and to simplify the operation.
The culture medium in the external mixing bag, homogenized by different agitation methods, flows between the matrix vessel in the incubator and mixing bag by the switch-over of negative / positive air pressures, submerging the matrices where cells attach. As a result, the matrices are submerged and exposed to air periodically. During the submerging phase, fresh nutrients replenish, and metabolic wastes are diffused and then carried away. Meanwhile, the carbon dioxide is forced to exhaust. During aeration, clean air is introduced and contacts with the porous matrices through a thin liquid film. The oxygen transfer efficiency is maximized. The culture medium is re-mixed and tuning for pH/DO in the mixing bag.
