TideCell Cell Harvest System (TCCHS) is specially designed for TideCell HD Cell Culture systems for cell recovery from the
cultured Matrix Vessel. The TCCHS is a powerful tool to harvest and transfer viable cells from large scale TideCell systems in a
close and automatic system. It can also be applied to harvest cell mass for biological production and any related applications.
Up to 1011 cells can be harvested within one to three hours operation. Entire cell harvest process is done in a close system and
could eliminate the risks of contamination.

Harvesting sufficient quantity of cells necessary for subsequent purification or for scale-up cell culture is labor-intensive yet critical task. To achieve higher consistency and efficiency for cell collection, it may require different maneuvers (longer trypsinization time, or relative gentle agitation) for multiple cell lines. However, the significant involvement of labor and various operation parameters applied may introduce variability to the manufacturing process or routine use.
 
TideCell Cell Harvest System is an automated workstation for cell detachment and collection in 2 L, 10 L, and 20 L TideCell matrix vessel. Cells can be harvested by the robotics from the matrix vessel, and proceed to the next application, for example, scale-up cell culture. The design for closed system operation could minimize the risk of contamination during the process. Large amounts of cells with high viability could be harvested within a short period of time and cell activities can be retained. The 6 peristaltic pumps with various reagent tanks, two gantry motors (shake and rotary motors) provide more control flexibility, and the user-friendly software allows the user to adapt a bench protocol, optimize the harvesting efficiency, and build up the standard procedures.
 
Parameters such as shake speed, rotation speed, incubation time, wash time, repeat cycles, and etc can be easily modified to fit the user’s specific demand.

Principle:
The whole procedure is based on the following steps:
1.          Wash of the adherent cells with PBS (removal of serum-containing medium and cell debris)
2.          Dissociation of cells from matrices with trypsin-EDTA solution (disruption of cell-substratum and cell-cell interactions)
3.          Inactivation of trypsin using inhibitors (termination of proteolysis process)
4.          Dispersion and harvesting of cells into the harvest tank (passaging of cells)
5.          Exhaust of the waste
